A Method of “Scope” PCR Abbreviated by Self-Complementation of Primers 5'end

Abstract

Scope (-fluorescent PCR) is the abbreviation of self-complementation of primers 5'end. A pair of 5'ends of primers with a central homologous bases is preceded by a complementary sequence with the 5' ends of the opposite primer, and a restriction endonuclease sequence is formed between the two. Now the new 5' ends of primer pairs with two partial homology segments is complemented to each other. So the 3'terminal complementation of the amplified products can also be template and mutually primer, and amplified by>2n index or to compensate for the consumption of primers. The sensitivity of Scope fluorescent PCR can be increased by 5.5 cycles in parallel, and there is no primer-dimer non-specificity because of the two homology segments.
